ECOHAB: Kamykowski_D- Florida Process Cruise: Karenia brevis counts, biochemistry and behavior, 1998-11-16 to 1998-11-19 (NCEI Accession 0000533)

Water bottle samples were collected from selected depths (variable with station) at 44 stations in the study area. Coulter counts in the 14 to 28 um size class were routinely determined for comparison with the microscope counts provided by other researchers on the cruise. At six stations, water samples were filtered through a 40 um Nytex mesh and centrifuged for 10 minutes at 1200 x g and then frozen to isolate Karenia brevis cells for subsequent analysis (pg cell-1) of cellular lipid, free amino acid, and protein. The biochemical determinations were performed within two weeks of collection at Kamykowski's laboratory at NCSU.

The biochemical analyses from the 1998 field program were based on relatively low K. brevis population numbers and are therefore at the lower end of the sensitivity of the biochemical techniques.

ECOHAB (ECology and Oceanography of Harmful Algal Blooms) is a scientific program designed to increase understanding of the fundamental processes underlying the impacts and population dynamics of HABs. This program addresses the many factors at organismal, population, community, and ecosystem levels that determine how HAB species respond to, and potentially alter their environment, the manner in which HAB species affect food-web and community interactions. The program is based on the results of a workshop co-sponsored by National Oceanic and Atmospheric Administration (NOAA) and National Science Foundation (NSF) and currently receives funding from several federal partners including NOAA, NSF, the U.S. Navy and the U.S. Environmental Protection Agency (EPA).
Purpose To verify that appropriately constrained Coulter counts can be used to monitor the abundance of Karenia brevis in natural water columns and that biochemical analyses of dense natural population of Karenia can provide interpretable data on the biochemical composition.
